ABSTRACT

At present, there is a concern about the quality of milk and diseases related to its consumption, as it can generate discomfort and allergic reactions in some individuals due to its protein components. Thus, the present study was developed to identify the allele and genotype frequencies of genes for ß-casein, A1 and A2, in dairy herds in the region of Araguaína-TO, Brazil. Genetic material from 421 animals (crossbred dairy cattle in lactation) was used. All animals were numbered for identification, and DNA samples were extracted from hair bulbs. Samples for two markers from the polymorphic regions were characterized and confirmed by real-time PCR using the ABI Prism® 7500 Sequence Detection System (Applied Biosystems). Allele and genotype frequencies were determined using the TaqMan™ detection system, where the primer and probe release different fluorescence signals for each allele of the polymorphism. The sampled herd showed frequencies of 28.27% for the A1 allele and 71.73% for the A2 allele. Genotype frequencies were 52.96% (223/421) for A2A2; 37.53% (158/421) for the A1A2 genotype; and 9.50% (40/421) for the A1A1 genotype. The frequency of the A1 allele for ß-casein in dairy herds from the northern region of Tocantins was low and is per the results of previous studies. Although the A2A2 genotype of ß-casein had a high relative frequency, the A1A2 genotype is still rather frequent, warranting greater selection pressure.(AU)


Atualmente existe uma preocupação em relação à qualidade e doenças que estão relacionadas ao consumo de leite, pois o mesmo pode gerar desconfortos e reações alergicas em alguns indivíduos devido aos seus constituintes protéicos. Assim, o presente estudo teve como objetivo identificar a frequência alélica e genotípica de genes para beta caseína, A1 e A2, em rebanhos leiteiros da região de Araguaína-TO. Foram utilizados material genético de 421 animais (bovinos leiteiros mestiços em lactação), e todos os animais foram numerados para identificação e amostras de DNA foram extraídas de bulbo de folículos pilosos. As amostras para dois marcadores das regiões polimórficas foram caracterizadas e confirmadas por PCR em tempo real, usando um sistema de detecção de sequências ABI Prism® 7500 (Applied Biosystems). As frequências alélicas e genotípicas foram determinadas utilizando o sistema de detecção TaqMan ™, no qual o primer e a sonda emitem diferentes sinais de fluorescência para cada alelo do polimorfismo. Observou-se frequência do alelo A1 de 28,27%, e do alelo A2 de 71,73% no rebanho amostral. A frequência genotípica de A2A2 foi de 52,96% (223/421), com genótipo A1A2 de 37,53% (158/421), e de 9,50% (40/421) animais com genótipo A1A1. A frequência do alelo A1 para beta-caseína em rebanhos leiteiros da região norte do Tocantins foi baixa e seguiu a mesma tendência já observada em estudos anteriores. Os genótipos A2A2 da beta-caseína apresentaram frequência relativa alta, entretanto o genótipo A1A2 ainda é bastante frequente, necessitando de maior pressão de seleção.(AU)

ABSTRACT

La prevalencia de trastorno del espectro autista ha ido en aumento, sin embargo, en Chile no existen lineamientos nutricionales acerca del posible tratamiento de la condición. Es por ello que el objetivo de esta revisión fue analizar la evidencia actual en relación al uso de una dieta libre de gluten y caseína, suplementación de vitamina D y omega 3 y su impacto en el comportamiento de niños/as con trastorno del espectro autista. Hay evidencia con resultados en torno a los beneficios de la suplementación con vitamina D debido a su carácter neuroprotector y su función neuromuscular. A su vez, la evidencia con omega 3 (DHA) es estadísticamente significativa para irritabilidad, hiperactividad, letargo, comportamiento estereotipado, conciencia social, comunicación y disminución de la severidad del autismo. Respecto a la dieta libre de gluten y caseína lo observado es que no existe evidencia que respalde los beneficios que esta exclusión entregaría. Aún falta evidencia para declarar un manejo nutricional específico para el tratamiento de los síntomas gastrointestinales y de comportamiento, más allá de la suplementación con aquellos micronutrientes en déficit.


The prevalence rate of autism spectrum disorder has been increasing, however, in Chile there are no nutritional guidelines about the possible treatment of the condition. That is the reason why the aim of this review is to analyze the current evidence regarding the use of a gluten and casein free diet, vitamin D and omega 3 supplementation and its impact in the behavior of children's with spectrum disorder autistic. There is evidence based on significant results regarding the benefits of vitamin D supplementation due to its neuroprotective character and neuromuscular function. At the same time the omega 3 evidence is statistically significant in the diminution of irritability hyperactivity, lethargy, stereotypical behavior, severity of autism and increase of social consciousness and communication. Regarding the gluten-free and casein-free diet, what was observed is that there is no evidence to support the benefits that this exclusion would provide. The evidence has not been conclusive to declare a specific nutritional management for the treatment of gastrointestinal and behavioral symptoms, beyond supplementation with those micronutrients in deficit.

ABSTRACT

Abstract Casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) complexes are anticariogenic and capable of remineralizing the early stages of enamel lesions. The use of fluoride prevents dental decay and the association of CPP-ACP with fluoride can increase remineralization. Objective: To evaluate the effect of CPP-ACP and CPP-ACPF creams associated with a fluoride dentifrice to prevent enamel demineralization in a pH cyclic model. Material and Methods: Previously selected by surface microhardness (SH) analysis, human enamel blocks (n = 56) were submitted to daily treatment with dentifrice in a pH-cycling model. The enamel blocks were divided into four groups; G1: Crest™ Cavity Protection - Procter & Gamble (1,100 ppmF of NaF); G2: Crest™ +MI Paste (MP) - Recaldent™ GC Corporation Tokyo, Japan); G3: Crest™ + MI Paste Plus (MPP) - Recaldent™ 900 ppm as NaF, GC Corporation Tokyo, Japan), and G4: control, saliva. Specimens were soaked alternatively in a demineralizing solution and in artificial saliva for 5 d. The fluoride dentifrice, with proportion of 1:3 (w/w), was applied three times for 60 s after the remineralization period. The undiluted MP and MPP creams were applied for 3 m/d. After cycling, SH was re-measured and cross section microhardness measurements were taken. Results: The SH values observed for the groups G3 (257±70), G1 (205±70), and G2 (208±84) differed from the G4 group (98±110) (one-way ANOVA and Tukey's post hoc test). There were no differences between the groups G1xG2, G2xG3, and G1xG3 for demineralization inhibition. The percentage of volume mineral showed that, when applied with fluoride dentifrice, MPP was the most effective in preventing enamel demineralization at 50 µ from the outer enamel surface (Kruskal-Wallis and Mann Whitney p<0.05). Conclusion: Fluoride dentifrice associated with CPP-ACPF inhibited subsurface enamel demineralization.

ABSTRACT

Metabolic acidosis is a cause of renal disease progression, and alkali therapy ameliorates its progression. However, there are few reports on the role of renal acid-base transporters during alkali therapy. We evaluated the effect of sodium bicarbonate therapy and the role of acid-base transporters on renal disease progression in rats with a remnant kidney. Sprague-Dawley rats consumed dietary sodium bicarbonate (NaHCO3) or sodium chloride (NaCl) with 20% casein after a 5/6 nephrectomy. After being provided with a casein diet, the NaHCO3-treated group had higher levels of serum bicarbonate than the control group. At week 4, the glomerular filtration rate in the NaHCO3 group was higher than that in the NaCl group, and the difference became prominent at week 10. The glomerulosclerosis and tubulointerstitial damage indices in the NaHCO3 group were less severe compared with controls at week 4 and 10. The expression of the Na/H exchanger (NHE) was decreased, and apical reactivity was decreased in the NaHCO3 group, compared with the NaCl group. Endothelin-1 levels in the kidney were also decreased in the NaHCO3 group. Dietary sodium bicarbonate has the effects of ameliorating renal disease progression, which may be related to the altered expression of NHE in the remaining kidney.

ABSTRACT

PURPOSE: To evaluate whether the prolonged consumption of flaxseed minimize the factors that trigger MS in healthy rats. METHODS: Pregnant rats were divided immediately after delivery into two groups during the lactation period, a control group (CG) receiving casein-based diet with 17% of protein, and a Flaxseed group (FG) with casein-based diet plus 25% of flaxseed. At weaning, 12 offspring of each group continued to receive the same feed but with 10% of protein up to 200 days old. RESULTS: FG showed a significant reduction in body weight (p=0.001), total cholesterol levels (p<0.0001), triglycerides (p=0.0001), and glucose (p=0.001). CONCLUSION: The flaxseed alters the indicators related to development of metabolic syndrome, because it has beneficial effects on lipids and glucose profiles and prevents the excess of body weight gain.


OBJETIVO: Avaliar se o consumo prolongado de linhaça pode minimizar os fatores que desencadeiam síndrome metabólica em ratos fêmeas saudáveis. MÉTODOS: Ratos fêmeas grávidas foram distribuidas após o parto em dois grupos durante o período de lactação: grupo controle (GC) recebendo dieta à base de caseína com 17% de proteína, e um grupo Linhaça (GL), com dieta à base de caseína adicionada de 25% de semente de linhaça. Ao desmame, 12 filhotes de cada grupo continuaram a receber as rações citadas acima, porém com 10% de proteína, até os 200 dias de vida. RESULTADOS: O GL mostrou uma redução significativa do peso corporal (p=0,001), no colesterol total (p<0,0001), triglicerídeos (p=0,0001) e na glicemia (p=0,001). CONCLUSÃO: A linhaça altera os indicadores relacionados ao desenvolvimento da síndrome metabólica por ter efeitos benéficos no perfil lipídico e glicêmico, e previne o excesso de ganho de peso corporal.

Introduction: Remineralization as a treatment procedure has received a lot of attention both from clinicians as well researchers. The objective of this in vitro study was to find out the efficacy of casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) and casein phosphopeptide-amorphous calcium phosphate fluoride (CPP-ACPF) in remineralizing enamel surface on which artificial caries lesion had been created. The changes were analyzed using DIAGNOdent® (KaVo) and scanning electron microscope (SEM). Materials and Methods: Ninety maxillary premolars were selected and divided into three groups of 30 teeth each: A (artificial saliva), B (CPP-ACP), and C (CPP-ACPF). All the samples were assessed using DIAGNOdent® at the baseline and after demineralization and remineralization. Three samples were randomly selected from each group after remineralization for surface evaluation using SEM. Results: Statistical analysis showed that group B {CPP-ACP (4.1±1.8)} and group C {CPP-ACPF (4.8±1.2)} had a significantly higher amount of remineralization than group A (1.7±0.7). Conclusion: All the three groups showed a statistically significant amount of remineralization. However, because of the added benefit of fluoride (NaF 0.2%), CPP-ACPF (Tooth Mousse-Plus®) showed marginally more amount of remineralization than CPP-ACP (Tooth Mousse®).

En el presente trabajo se evaluó el efecto hiperlipídico de una dieta combinada de caseína y grasas saturadas, en un modelo biológico conformado por ratas Wistar. Se integraron tres grupos experimentales de seis animales cada uno: el primero (grupo control) consumió una dieta convencional; el segundo, una dieta convencional combinada con caseína y grasa de cerdo al 20 por ciento y el tercero, la dieta convencional suplementada solamente con grasa de cerdo al por ciento. Fueron cuantificados el colesterol y los triacilglicéridos por métodos enzimáticos en tres momentos diferentes: al comienzo del estudio, a los 15 días y al finalizar este. El incremento de peso corporal, una vez finalizada la investigación, no mostró diferencias significativas entre los grupos de experimentación. El grupo de la dieta combinada elevó el perfil lipídico más eficientemente que el que solo consumió grasas saturadas, lo cual confirma que el modelo experimental de hiperlipidemia inducida por dieta suplementada con caseína y grasas saturadas es el más eficaz para la elevación del perfil lipídico


In the current research it was evaluated the hyperlipidic effect of a combined diet with casein andsaturated fats, in a biological model conformed by Wistar's rats. Three experimental groups wereintegrated with six animals in each one. The first group (control group) consumed a conventionaldiet; the second group, a conventional diet combined with casein and pig fat at 20 %; and the thirdone, the same conventional diet supplemented just with pig fat at 20 %.The cholesterol andtriacylglycerols were quantified by means of enzymatic methods in three different moments: at thebeginning of the study, after 15 days and at the end of this. The increase of body weight, once itwas finished the research, didn't show significant differences among the experimental groups. Thegroup of the combined diet increased its lipidic profile more efficiently than the group that justconsumed saturated fats, which confirms that the experimental model of induced hyperlipidaemiaby supplemented diet with casein and saturated fats is the most effective for the elevation of lipidicprofile.

The flavonoid rutin is recognized as playing an important role in the protection of plants against lepidopterans. Bioassays with this compound are generally carried out using artificial diets. Proteins of high energy value, such as casein, are important ingredients of insect artificial diets as a source of essential amino acids. However, such proteins can generally increase the allelochemical activity. Our objective was to evaluate the effects of rutin on larvae of the velvetbean caterpillar Anticarsia gemmatalis Hübner by incorporating this allelochemical into diets with different concentrations of casein. Three casein concentrations (0, 7 g, or 14 g) combined with none, 0.65 percent, or 1.30 percent of rutin were added to the rearing diet and offered to the larvae from hatching to pupation. Rutin negatively affected larval development, the amount of food consumed, and pupal weight of A. gemmatalis. These negative effects were clearly seen in insects fed on diets with 7 g of casein to which any concentration of rutin was added. The effects of rutin when added to the diets without casein were stronger than in diets containing a suitable amount of casein (14 g). The greater negative effects of rutin in diets containing suboptimal concentrations of casein indicate that casein can increase the effects of rutin only when the diets are nutritionally unsuitable for insect development.

The purpose of this study was to analyze the influence of diets varying in lipids and proteins on the histopathologic variety of murine salivary tumors induced by DMBA. 117 BALB/c mice were assigned to experiments one (E1: lipids, males) and two (E2: proteins, males and females), E1 comprising Soy oil (SO); Corn oil (CO, control); Fish oil (FO) and Olein (O) groups and E2, soy protein (SP) and casein (C) groups. Tumors were induced by DMBA and the animals were sacrificed at week 13- post-induction. Tumor volume was calculated. Tumor sections were stained with H-E for histopathologic evaluation. No significant association was found between tumor volume and dietary condition (p > 0.05). In E1, FO animals developed mainly carcinomas (C) (58.8%), the sarcomas (S) and carcinosarcomas (CS) being especially of high-grade type (tumors < 600 mm3). In E2, SP animals developed mainly C (55.6%). Although no significantly different (p > 0.05), S and C were more frequent in female and male mice, respectively. In both E1 and E2, the biggest tumors (> 600 mm3) were mainly high-grade S (87.5%-80%). Dietary fat and soy protein appear to influence the tumor histopathology and thus its prognosis.


El objetivo de este estudio fue analizar la influencia de dietas con diferente contenido de lípidos y proteínas sobre la variedad histopatológica de tumores salivares murinos inducidos por DMBA. Se asignaron 117 ratones BALB/c a los experimentos uno (E1: lípidos, machos) y dos (E2: proteínas, machos y hembras). E1 comprendió a los grupos aceite de soja (AS), aceite de maíz (AM, control), aceite de pescado (AP) y oleína (O), en tanto E2 incluyó a los grupos preteína de soja (PS) y caseína (C). Los tumores fueron inducidos por DMBA y los animales fueron sacrificados a la 13ª semana post-inducción. Se calculó el volumen tumoral. Los cortes de tumor fueron coloreados con Hematoxilina-Eosina para su evaluación histopatológica. No se encontró asociación entre volumen tumoral y condición dietaria (p>0.05). En E1, los animales del grupo AP desarrollaron principales carcinomas (C) (58,8%), en tanto que los sarcomas (S) y carcinosarcomas (CS) fueron de alto grado (tumores<600 mm³). En el E2, los animales del grupo PS desarrollaron principalmente C (55.6%). Aunque la diferencia no fue significativa (p>0.05), S y C fueron más frecuentes en ratones hembras y machos, respectivamente. Tanto el E1 com en E2, los tumores más voluminosos (> 600 mm³) fueron principalmente de alto grado (87.5%-80%) Los lípidos y la proteína de soja de la dieta parecen influenciar la histopatología de los tumores y, en consecuencia, su pronóstico.

Se presenta un caso clínico piloto, en el marco de un protocolo de investigación clínica sobre remineralización de las manchas blancas. Se indican dos tipos de tratamientos efectuados, con y sin grabado previo del esmalte con ácido fosfórico al 35-42 por ciento y la aplicación de una pasta remineralizante a base de fosfopéptidos de la caseína/fosfato de calcio amorfo (MI Paste, Recaldent, G.C., Japan); complementada con la administración de goma de mascar con los mismos componentes (Trident, Recaldent, Adams). A los 60 días, se observó un notorio cambio de color en la zona desmineralizada grabada previamente con ácido, con respecto a la que no fuera grabada, indicando la maejor penetración del producto remineralizante al abrir los poros del esmalte desmineralizado

Most contacts with food protein and microbiota antigens occur at the level of the gut mucosa. In animal models where this natural stimulation is absent, such as germ-free and antigen-free mice, the gut-associated lymphoid tissue (GALT) and systemic immunological activities are underdeveloped. We have shown that food proteins play a critical role in the full development of the immune system. C57BL/6 mice weaned to a diet in which intact proteins are replaced by equivalent amounts of amino acids (Aa diet) have a poorly developed GALT as well as low levels of serum immunoglobulins (total Ig, IgG, and IgA, but not IgM). In the present study, we evaluated whether the introduction of a protein-containing diet in 10 adult Aa-fed C57BL/6 mice could restore their immunoglobulin levels and whether this recovery was dependent on the amount of dietary protein. After the introduction of a casein-containing diet, Aa-fed mice presented a fast recovery (after 7 days) of secretory IgA (from 0.33 to 0.75 mg/mL, while in casein-fed mice this value was 0.81 mg/mL) and serum immunoglobulin levels (from 5.39 to 10.25 mg/mL of total Ig). Five percent dietary casein was enough to promote the restoration of secretory IgA and serum immunoglobulin levels to a normal range after 30 days feeding casein diet (as in casein-fed mice - 15 percent by weight of diet). These data suggest that the defect detected in the immunoglobulin levels was a reversible result of the absence of food proteins as an antigenic stimulus. They also indicate that the deleterious consequences of malnutrition at an early age for some immune functions may be restored by therapeutic intervention later in life.

Patients residing in endemic areas for schistosomiasis in Brazil are usually undernourished and when they develop the hepatosplenic clinical form of the disease should usually receive hospital care, many of them being in need of nutritional rehabilitation before specific treatment can be undertaken. In the mouse model, investigations carried out in our laboratory detected a reduced aminoacid uptake in undernourished animals which is aggravated by a superimposed infection with Schistosoma mansoni. However, in well-nourished infected mice no dysfunction occurs. In this study, we tried to improve the absorptive intestinal performance of undernourished mice infected with S. mansoni by feeding them with hydrolysed casein instead of whole casein. The values obtained for the coefficient of protein intestinal absorption (cpia) among well-nourished mice were above 90 percent (either hydrolysed or whole protein). In undernourished infected mice, however, the cpia improved significantly after feeding them with hydrolysed casein, animals reaching values close to those obtained in well-nourished infected mice

ABSTRACT

OBJECTIVE: To assess the effect of malnutrition on the development of giardiasis in Sprague-Dawley rats, using different inoculum sizes of Giardia lamblia cysts. MATERIAL AND METHODS: An experimental study was conducted between 1995 and 1999 at Centro de Investigación, Alimentación y Desarrollo (Center for Research, Food, and Development), in Hermosillo, Sonora, Mexico. The study population consisted of two groups of six to eight experimental units that were fed two different diets and inoculated five different concentrations of Giardia lamblia cysts. Data were collected on excretion of cysts, weight gain, food intake, bowel contents, and macro and microscopic lesions in the intestinal mucosa. Statistical analysis consisted of analysis of variance and residuals. RESULTS: Animals fed with a diet meeting nutritional requirements required an infecting inoculum of 60 cysts, while malnourished rats required only six cysts to develop mucosal lesions. CONCLUSIONS: Weight gain monitored during ten days was not a good indicator of Giardia lamblia infection. Infection depended on cyst inoculum size as well as on the nutritional status of the tested animals.

Adult male rats received daily injections (sc) of gonadotropin releasing hormone antagonist (0.2 mg/kg(-1) x day(-1)) for 21 days when they were sacrificed on day 22, adrenal weight, adrenal A5-3beta (delta 5-3beta) hydroxysteroid dehydrogenase (Delta5-3beta-HSD) activity and serum level of corticosterone were increased significantly while testicular 17beta (17beta) hydroxysteroid dehydrogenase (17beta-HSD) activity and serum level of testosterone and spermatogenesis were decreased in the rats fed on 5% casein diet. GnRH antagonist treated rats fed on 20% casein diet, resulted significant decrease in adrenal weight, serum corticosterone and adrenal A5-3beta-HSD activity while testicular 17beta-HSD activity serum testosterone levels and the weights of sex organs were increased with respect to anti GnRH treated rats fed on 5% casein diet. But the GnRH antagonist treated rats fed on 20% casein diet showed decreased spermatogenesis quantitatively and sperm count appeared similar to anti GnRH treated rats fed on 5% casein diet. These results indicate that high casein diet protects adrenocortical activity and stimulates testosterone synthesis without effecting spermatogenic arrest in GnRH antagonist treated rats. It may be concluded that GnRH antagonist in presence of high milk protein diet may be considered to be a suitable antihormone in the development of an ideal male contraceptive.

ABSTRACT

El propósito del presente trabajo fue estudiar el efecto de la rehabilitación nutricia con dietas que contenían diferente concentración de proteína (6, 18 o 50 por ciento) en ratas previamente desnutridas sobre los niveles del factor de crecimiento semejante a la insulina (IGF-I) y de las proteínas que unen al factor semejante a la insulina (IGFBPs). La desnutrición se indujo alimentando ratas en crecimiento con 0.5 por ciento de caseína por cinco semanas. En este periodo, el crecimiento, las proteínas totales en el suero, las concentraciones de IGF-I en suero y la relación IGFBP-3/IGFBP-2 se encontraron significativamente disminuidas comparados con el grupo con libre acceso a una dieta con 18 por ciento de caseína. La rehabilitación nutricia por 21 días con 6 por ciento de caseína no cambió ninguno de estos parámetros. La rehabilitación nutricia con una dieta con 18 o 50 por ciento de caseína por un día no inició el restablecimiento de las concentraciones de IGF-I y la relación IGFBP-3/IGFBP-2. Sin embargo, después de 10 días con dietas al 18 o 50 por ciento de caseína por un día no inició el restablecimiento de las concentraciones de IGF-I y la relación IGFBP-3/IGFBP-2. Sin embargo, después de 10 días con dietas al 18 o 50 por ciento de caseína, las ratas aumentaron 12 veces las concentraciones de IGF-I y siete veces la relación IGFBP-3/IGFBP-2. Finalmente, la rehabilitación por 21 días con 18 o 50 por ciento de caseína produjo un incremento de 21 y 16 veces en las concentraciones de IGF-I y 6.1 y 14.5 veces en la relación IGFBP-3/IGFBP-2 respectivamente. Estos resultados sugieren que la rehabilitación nutricia con 18 por ciento o 50 por ciento de caseína son más efectivas que dietas que contienen 6 por ciento de caseína para restablecer el peso corporal. Las concentraciones de IGF-I e IGFBP-3 fueron indicadores sensibles de la evolución del estado nutricio dependiendo de la concentración de proteína en la dieta

ABSTRACT

Se estudió el efecto de la concentración de la proteína de la dieta sobre concentraciones de ARNm de la tirosina aminotransferasa (TAT) y la fenilalanina hidroxilasa (PAH) hepáticas en ratas adaptadas a consumir dietas con 18 ó 50 por ciento de caseína en un horario restringido de 7 horas (9 a 16 h) durante 5 días. Las concentraciones de ARNm de TAT de ratas adaptadas a una dieta de 18 por ciento de caseína y alimentadas en forma aguda con dietas que contenían 6, 18 ó 50 por ciento de caseína, fueron 0.15, 0.84 y 5.08 veces más altas a las 6 horas en comparación con las concentraciones de ARNm antes de la administración de la dieta. Las concentraciones de ARNm de TAT después de 17 horas de ayuno en las ratas alimentadas con 6, 18 ó 50 por ciento de caseína fueron respectivamente -0.45, 1.76 y 9.11 veces mayores en comparación con el valor basal. Las concentraciones ARNm de PAH mostraron un patrón similar; en las ratas adaptadas a 18 por ciento de caseína se observó un aumento de -.68, 1.63 y 2.5 veces en las concentraciones de ARNm de PAH en las ratas alimentadas en forma aguda con 6, 18 y 50 por ciento de casína respectivmanete y un aumento de -0.86, 2.32 y 9.33 veces después de 17 horas de ayuno. La concentraciones de ARNm de TAT y PAH en ratas adaptadas a consumir 50 por ciento de caseína y luego alimentadas con 6 ó 50 por ciento de caseína mostraron un pico máximo a las 6 horas de ayuno. Estos resultados sugieren que las concentraciones crecientes de proteína en la dieta son capaces de producir aumentos en la concentración de los ARNm de las dos enzimas, posiblemente para eliminar el exceso de aminoácidos consumidos, ya que la concentración de los ARNm dependió más del contenido de proteína de la dieta de adaptación

ABSTRACT

Estudios previos, realizados por Hevia y colaboradores en 1980, mostraron que en ratas la adición de 5 por ciento de L-lisina una dieta en base a caseína producía, a los 15 días, una disminución en la velocidad de crecimiento de los animales y una acumulación marcada de lípidos en el hígado, principalmente de triglicéridos y colesterol. Debido a que la lisina es un aminoácido esencial utilizado en la fortificación de alimentos, el presente trabajo determinó si el consumo de concentraciones de lisina similares a las utilizadas en la fortificación de alimentos para consumo humano, las cuales abarcan un rango comprendido entre 0,1 y 0,25 gramos por cada 100 gramos de alimento, producen alteraciones similares a las antes mencionadas. Para ello se utilizaron ratas en crecimiento de la cepa "Sprague-dawley", las cuales fueron sometidas a diferentes tratamientos a base de dietas conteniendo 15 por ciento de proteína proveniente de la caseína-maíz (2:1) y suplementadas con 0,1; 0,25 y 5 por ciento de L-lisina durante 15, 45 o 60 días. Los resultados mostraron que, a los 15 días de tratamiento, el grupo de animales alimentados con 15 por ciento de caseína y 5 por ciento de lisina presentó además de una marcada reducción en la velocidad de crecimiento, un hígado graso caracterizado principalmente por una acumulación severa de triglicéridos y colesterol. Similares resultados se obtuvieron con el grupo de animales sometidos a una dieta en base a 15 por ciento de proteína proveniente de la mezcla caseína-maíz suplementada con 5 por ciento de L-lisina, aunque la acumulación de lípidos hepáticos fue de menor magnitud. Por otra parte, a diferencia de los resultados obtenidos de los grupos antes mencionados, aquellos que consumieron concentraciones menores de lisina dietaria, no presentaron alteraciones en el metabolismo lipídico a los 15 días de tratamiento, observándose además un crecimiento acelerado y significativamente mayor al obtenido por los grupos que no consumieron lisina. Al prolongar el tiempo de tratamiento hasta 45 días se observó por una parte, la reservación del hígado graso ocasionado por el consumo de 5 por ciento de lisina dietaria y un incremento significativo en los lípidos circulares, principalmente de triglicéridos y colesterol y, por otra parte, un aumento progresivo de la concentración de lípidos hepáticos en aquellos grupos alimentarios con cantidades menores de lisina en la dieta

ABSTRACT

The purpose of this study was to determine whether the addition of high levels of years to casein and soy diets could modify the well known effects of any of these proteins on plasma cholesterol. Rabbits, were fed either a diet containing soybean protein-brewer's yeast or casein-brewer's yeast (each protein source providing 50 per cent of the dietary nitrogen content) and casein and soybean protein basal diets. Brewer's yeast was obtained from a local beer factory in its non-debitered form. The diets contained 20 per cent protein, 9 per cent coconut oil and 1 per cent corn oil, with no added cholesterol. After a 22 day experimental period, rabbits fed casein developed hypercholesterolemia whereas those fed the soy bean protein diet did not. The replacement of 50 per cent of hte soy nitrogen by brewer's yeast nitrogen, increased the total cholesterol plasma level, but significant differences were only observed between rabbits fed casein and casein-yeast and those fed soybean protein. No differences in high density lipoprotein cholesterol could be detected among the groups. However, the HDL-cholesterol/total cholesterol ratio was significantly reduced in responce to soy substitution by brewer's yeast. The (low density lipoprotein + very low density lipoprotein)-cholesterol was increased in all groups whith the exception of the animals fed purely soy protein. these data suggest a hypercholesterolemic activity of the dietary non-debittered brewer's yeast. Nevertheless, according to the amino acid composition, the factor responsible for the reported effects of dietary yeast was not associated with a high lysine to arginine ratio which could be due to extracellular components

ABSTRACT

1. In order to study the effects of protein quantity and quality on development, Wistar rats were submitted to four different diets during lactation and post-lactation periods. Three isocaloric diets were utilized with 6 per cent (M), 16 per cent (W16) and 20 per cent (W20) of protein (casein), and the fourth diet (C) consisted of a commercial lab chow containing 22 per cent protein. 2. During the lactation and post-lactation periods the body weights of dams and pups were recorded weekly. On the 49th day of age (21 days of suckling and 28 days of ad libitum chow), all animals were sacrificed and the brains removed and weighed. 3. Dams from all groups increased food intake during the lactation period, but dams of the M group consumed a lower amount of diet as compared with other groups. Only the body weight of M dams was affected by diet during the lactation period, but the body weight of pups from the M and C groups was lower than in the other two groups. At 49 days of age C and M pups showed a significantly lower brain weight compared with W16 and W20 pups. 4. Thus, a commercial lab chow diet does not promote normal body and brain development as compared with balanced diets containing 16 or 20 per cent protein. These results emphasize the need for further studies in order to evaluate other biological and behavioral parameters that might be altered by a lab chow diet
